Abstract
Enterotoxigenic Bacteroides fragilis (ETBF) cause diarrhea and are implicated in inflammatory bowel diseases (IBD) and colorectal cancer. However, up to 30% of asymptomatic individuals harbor ETBF. The only known virulence factor for ETBF is the Bacteroides fragilis toxin (BFT) which induces E-cadherin cleavage, activation of the beta-catenin pathway and epithelial cell proliferation. We investigated whether ETBF could promote colonic tumor development in mice genetically predisposed to develop intestinal tumors (Min, multiple intestinal neoplasia). We found that ETBF colonization triggers colonic inflammation and strongly induces colonic tumors in Min mice. ETBF induced STAT3 activation in the colon and a dominant IL17 response in TCRαβ+CD4+ and TCRγδ+CD4-CD8- T cells. Antibody neutralization experiments using anti-IL17 and anti-IL23R antibodies significantly inhibited ETBF-induced colitis and colonic tumor formation. These results suggest ETBF may promote colonic tumor development via the IL17/IL23 pathway.
