Evaluating the Role of Cellular Prion Protein (PrPC) in Self-renewal and Differentiation of Human Embryonic Stem Cells for Future Clinical Applications

The normal, cellular isoform of the prion protein, PrPC is a sialoglycoprotein that is attached to the cell membrane via a C-terminal glycosylphosphatidyl-inositol (GPI) anchor and regarded as an essential partner of prion diseases. The spectrum of proposed biological functions of PrPC has been expanding rapidly over the last decade and several studies illustrated that the involvement of PrPC in stem cell proliferation and differentiation is broader than that we have thought. Here we have employed human embryonic stem cells (hESCs) for assessing the role of PrPC in human embryogenesis and development and showed that ectopic expression of PrPC in hESCs under self-renewal conditions triggered differentiation toward lineages of three germ layers. Silencing of PrPC under spontaneous differentiation conditions suppressed differentiation of cells toward ectodermal lineages, but did not affect differentiation of endodermal and mesodermal lineages, thereby changing the balance between the lineages of three germ layers. Taken together, these data support the appearance of new strategies to control stemness, which could represent important advances in the field of regenerative medicine.
