A novel role of Prx1 as an RNA-binding protein: Prx1 as a post-transcriptional regulator of snoRNAs

Prx1 is a member of the peroxiredoxin (Prx) family that detoxifies various peroxide substrates through conserved catalytic Cys residues. Prx1 mediates redox signaling by altering protein-protein interactions. For example, Prx1 interacts with MAPK phosphatases, thereby regulating breast cancer cell senescence by specifically coordinating p38 MAPK-induced signaling in a H2O2-dependent manner. Prx1 also interacts with other important signaling molecules, such as PTEN and NF-κB. In addition to the known roles of Prx1 in protein-protein interactions, we have recently shown that Prx1 binds to RNAs and has RNA chaperone activity in vitro. Our initial discovery of this novel function of Prx1 as a RNA-binding protein (RBP) was further supported by three studies in which Prx1 was identified as a RBP candidate through comprehensive analyses of the mRNA interactome in in HeLa cells, HEK293 cells, and embryonic stem cells. The discovery of Prx1 as a RBP has important biological implications. RBPs regulate RNA molecules through RNA processing, splicing, stability, localization, translation, and degradation. The deregulation of RBPs has been implicated in various diseases, ranging from neurologic disorders to cancer. Recent studies have shown that we can gain novel insights into the functional roles of RBPs by understanding RBP–RNA interactions. As multiple studies, including our initial discovery, support that Prx1 is an RBP, yet Prx1 function as an RBP remains unknown. To characterize the role of Prx1 as an RBP, we pulled down Prx1-RNA complexes and sequenced target RNAs of Prx1. Through sequencing and further validation studies, we found that Prx1 binds to s specific subset of small nucleolar RNAs (snoRNAs) and regulates these molecules at the post-transcriptional level. In this talk, I will discuss the novel role of Prx1 as a post-transcriptional regulator of snoRNAs.
