Abstract
Protein chip system has attracted a great interest as the next generation of biosensor. Protein chip systems consist of a sensor chip and a detection system. Several types of non-selective sensor chips has been developed by the use of various protein immobilizers including carboxymethyl-dextran, disulfide chemicals, and calix-crown molecules on very thin gold film. Currently, selective protein chip surfaces, such as hydrophobic, hydrophilic and metal binding surfaces, are under development in many research groups. Proteins bound on chip surfaces can be detected by detection systems using various methods, such as surface plasmon resonance (SPR), fluorescence and mass spectrometry.
We have developed selective protein chip surfaces using polymers with charged- active groups such as quaternary ammonium and sulfonate. Metal surfaces including Fe3+, Ni2+, Co2+ have also been applied to develop selective chip surfaces. Currently, protein binding studies are under intensive investigation. A detection system to automatically scan 100 protein spots (10x10, φ=1mm) on a chip has been developed using white lights. Currently, we are working on the increase of scanning speed and accuracy of protein detection. Protein chip system has various biological applications, such as screening of drugs and chemicals, kinetic measurement of protein-protein interaction, protein concentration measurement, protein purification, and protein characterization. In addition, the chip system can be used for enzyme activity assay and identification of disease markers. Thus, protein chip system is a new integrated system for biomolecule analysis in the Proteomic Era.
