Abstract

Analysis of the complete genomic structure of the human and mouse ribosomal protein S3 (rpS3) genes revealed the presence of a functional U15 snoRNA genes in its intron. The first and fifth introns of human and mouse ribosomal protein S3 genes appear to have functional U15 snoRNAs. Interestingly, extra-ribosomal function of rpS3 protein is reported. This protein is also known as UV DNA repair endonuclease III  because the protein encoded by human and/or mouse rpS3 gene shows an endonuclease activity on UV irradiated or oxidatively damaged DNAs. Therefore, rpS3 seems to function both in translation and in DNA repair. It is to be noted that the column profile of the endonuclease activity of rpS3 appears to be altered in Xeroderma Pigmentosum(XP) group D cells. XP is a human disease characterized by high sensitivity of skin by UV- or sun light irradiation and by high frequency of developing skin cancers. I am also reporting here a 3rd function of rpS3 protein which induces apoptosis after post-translational modification under stress conditions in a p53 independent manner. The mechanism of cell death appears to be mediated by caspases-8 and 3. N- and C-terminal domains of rpS3 are responsible for apoptosis and DNA repair, respectively.
